Autonomous Motion of Metallic Microrods Propelled by Ultrasound

Autonomously moving micro-objects, or micromotors, have attracted the attention of the scientific community over the past decade, but the incompatibility of phoretic motors with solutions of high ionic strength and the use of toxic fuels have limited their applications in biologically relevant media. In this letter we demonstrate that ultrasonic standing waves in the MHz frequency range can levitate, propel, rotate, align, and assemble metallic microrods (2 μm long and 330 nm diameter) in water as well as in solutions of high ionic strength. Metallic rods levitated to the midpoint plane of a cylindrical cell when the ultrasonic frequency was tuned to create a vertical standing wave. Fast axial motion of metallic microrods at ~200 μm/s was observed at the resonant frequency using continuous or pulsed ultrasound. Segmented metal rods (AuRu or AuPt) were propelled unidirectionally with one end (Ru or Pt, respectively) consistently forward. A self-acoustophoresis mechanism based on the shape asymmetry of the metallic rods is proposed to explain this axial propulsion. Metallic rods also aligned and self-assembled into long spinning chains, which in the case of bimetallic rods had a head-to-tail alternating structure. These chains formed ring or streak patterns in the levitation plane. The diameter or distance between streaks was roughly half the wavelength of the ultrasonic excitation. The ultrasonically driven movement of metallic rods was insensitive to the addition of salt to the solution, opening the possibility of driving and controlling metallic micromotors in biologically relevant media using ultrasound.

Acoustic Propulsion of Nanorod Motors Inside Living Cells

The ultrasonic propulsion of rod-shaped nanomotors inside living HeLa cells is demonstrated. These nanomotors (gold rods about 300 nm in diameter and about 3 mm long) attach strongly to the external surface of the cells, and are readily internalized by incubation with the cells for periods longer than 24 h. Once inside the cells, the nanorod motors can be activated by resonant ultrasound operating at 4 MHz, and show axial propulsion as well as spinning. The intracellular propulsion does not involve chemical fuels or high-power ultrasound and the HeLa cells remain viable. Ultrasonic propulsion of nanomotors may thus provide a new tool for probing the response of living cells to internal mechanical excitation, for controllably manipulating intracellular organelles, and for biomedical applications.

From One to Many: Dynamic Assembly and Collective Behavior of Self-Propelled Colloidal Motors

The assembly of complex structures from simpler, individual units is a hallmark of biology. Examples include the pairing of DNA strands, the assembly of protein chains into quaternary structures, the formation of tissues and organs from cells, and the self-organization of bacterial colonies, flocks of birds, and human beings in cities. While the individual behaviors of biomolecules, bacteria, birds, and humans are governed by relatively simple rules, groups assembled from many individuals exhibit complex collective behaviors and functions that do not exist in the absence of the hierarchically organized structure. Self-assembly is a familiar concept to chemists who study the formation and properties of monolayers, crystals, and supramolecular structures. In chemical self-assembly, disorder evolves to order as the system approaches equilibrium. In contrast, living assemblies are typically characterized by two additional features: (1) the system constantly dissipates energy and is not at thermodynamic equilibrium; (2) the structure is dynamic and can transform or disassemble in response to stimuli or changing conditions. To distinguish them from equilibrium self-assembled structures, living (or nonliving) assemblies of objects with these characteristics are referred to as active matter. In this Account, we focus on the powered assembly and collective behavior of self-propelled colloids. These nano- and microparticles, also called nano- and micromotors or microswimmers, autonomously convert energy available in the environment (in the form of chemical, electromagnetic, acoustic, or thermal energy) into mechanical motion. Collections of these colloids are a form of synthetic active matter. Because of the analogy to living swimmers of similar size such as bacteria, the dynamic interactions and collective behavior of self-propelled colloids are interesting in the context of understanding biological active matter and in the development of new applications. The progression from individual particle motion to pairwise interactions, and then to multiparticle behavior, can be studied systematically with colloidal particles. Colloidal particles are also amenable to designs (in terms of materials, shapes, and sizes) that are not readily available in, for example, microbial systems. We review here our efforts and those of other groups in studying these fundamental interactions and the collective behavior that emerges from them. Although this field is still very new, there are already unique and interesting applications in analysis, diagnostics, separations, and materials science that derive from our understanding of how powered colloids interact and assemble.

3D steerable, acoustically powered microswimmers for single-particle manipulation

The ability to precisely maneuver micro/nano objects in fluids in a contactless, biocompatible manner can enable innovative technologies and may have far-reaching impact in fields such as biology, chemical engineering, and nanotechnology. Here, we report a design for acoustically powered bubble-based microswimmers that are capable of autonomous motion in three dimensions and selectively transporting individual synthetic colloids and mammalian cells in a crowded group without labeling, surface modification, or effect on nearby objects. In contrast to previously reported microswimmers, their motion does not require operation at acoustic pressure nodes, enabling propulsion at low power and far from an ultrasonic transducer. In a megahertz acoustic field, the microswimmers are subject to two predominant forces: the secondary Bjerknes force and a locally generated acoustic streaming propulsive force. The combination of these two forces enables the microswimmers to independently swim on three dimensional boundaries or in free space under magnetical steering.
