Trastuzumab-deruxtecan (T-DXd) has demonstrated remarkable results in advanced breast cancer. However, 10-15% of treated patients develop interstitial lung disease (ILD), which is a frequent cause of treatment discontinuation. Potential germline markers in ILD susceptibility remain unexplored.
Tertiary lymphoid structures (TLS) drive adaptive antitumor immunity, impacting prognosis and immunotherapy efficacy across cancers. However, no established classification exists for TLS in breast cancer (BC). This study aims to define a novel hematoxylin and eosin (H&E)-based TLS scoring system in BC and identify gene expression-based biology indicative of TLS presence.
The Prosigna/PAM50 assay classifies early breast cancer (eBC) by intrinsic subtype and Risk of Recurrence (ROR), aiding in adjuvant treatment decisions. While treatment strategies are clear for low and high-risk groups, intermediate-risk cases require further clinical context integration. This real-world study aimed to estimate relapse-free survival (RFS) and identify clinical factors associated with the intermediate-risk group (i-ROR) that may influence treatment decisions.
