We have determined the sequence of a 2210-base pair DNA segment containing genes required for expression of Eco RI DNA restriction and modification phenotypes.Polypeptide sequences encoded within the two longest open reading frames in this region are in agreement with NH2-and COOH-terminal sequences of the two Eco RI enzymes (Rubin, R.
